What kind of buffer should be used to dilute the DNA samples?
The DNA should be diluted in 10mM Tris/1mM EDTA (TE buffer).
Which method should we use to determine DNA concentration?
Quantify DNA using a dsDNA-specific method such as PicoGreen (Invitrogen, Catalog #P7581) or Qubit.
What is the amount and volume of DNA required?
The total amount of DNA and total volume required is dependent on the particular assay requested. For Illumina Infinium microarray services, the minimum sample concentration is 50ng/ul and minimum sample volume is 10ul. All DNA samples must have identical volumes.
What is the requirement for DNA quality?
DNA must be intact, of high molecular weight and have an OD 260/280 of 1.8-2.0. DNA must be free of contaminating nucleic acids from other individuals or species.
What format should be used to send DNA samples?
DNA samples should be submitted in 96-well PCR plates and covered with heat sealer or ABI film. Include a sample manifest with a brief description of the DNA extraction protocols.
Does the PGC offer sample extraction and plating services for other sample types besides blood?
Yes, DNA sample extraction and plating services are available for other sample types such as saliva, tissue or FFPE samples. For more information, please contact the PGC manager.
