T-cell Receptor Beta (TCRB) Gene Rearrangement (PCR)

Indication

– Assessment of clonality in suspected T-cell lymphoproliferations and lymphoproliferative neoplasm of uncertain lineage
– As a marker of neoplastic cells in a subset of B- and T-lymphoblastic leukemias

Methodology

A PCR method involving fluorescent-labeled PCR products and capillary electrophoresis is performed to detect only complete TCRB rearrangements.

Test Parameters

The sensitivity of this assay for detection of a clonal T-cell population is between 1:100 and 1:1,000 lymphocytes, depending on the number of polyclonal T-cells present. T-cell tumor expressing the gamma-delta T-cell receptor will not be detected. Clonal TCRB rearrangements can be seen in B-cell acute lymphoblastic leukemias and are thus not definitive for lineage in immature lymphoid neoplasms.

• 10 ml peripheral blood in purple top tube (EDTA Vacutainer), sent on wet ice
