Identifying therapeutic targets to enhance radiation sensitivity for clinical translation
The goals of this theme are to identify druggable pathways to enhance radiation/chemoradiation effect on tumor cells and to identify therapeutic targets to reverse radiation resistance. To do so, we use high throughput clonogenic survival with genomic variants (KRAS, LKB1, KEAP1) and radiation resistance models.

The goals of this theme are to identify novel immune effectors of radiation therapy and therapeutic targets that would enhance immunologic response to radiation therapy.

Understanding radiation induced immune suppression for mitigation strategies
This theme involves understanding and modeling to predict radiation induced lymphopenia to improve radiation approaches (protons/photons). It deals with the development of drug-based mitigation strategies after chemoradiation to improve immunotherapy effectiveness.

Theme 4
Predictive biomarkers of treatment response to enable risk stratification for treatment intensification
This theme's goals are to identify tumor-microenvironmental factors involved in mediating CRT response and to assess the use of non-invasive imaging to predict treatment response for proper risk stratification of patients.
